I wanted to see if I could find additional proof of relationship to my Luxembourg ancestors and did the DNA test. It shows I have cousins who claim the same family tree. Would this help at all in proving a relationship, particularly where my great grandmother's birth certificate is missing?

I doubt it. At this point, DNA testing to ascertain countries of origin isn't an exact science.
Have you contacted the cousins? Maybe they know how to obtain a copy of your ancestor's birth certificate
